The second installment in Richelle Mead's Bloodlines series is set in a world where three types of vampires exist alongside humans: Moroi (living, magic-wielding vampires), dhampirs (half-human, half-vampire guardians), and Strigoi (evil, undead vampires). The Alchemists are a secret human society dedicated to keeping the vampire world hidden, using science, vampire blood-infused golden lily tattoos that each member wears as a sworn vow to their beliefs, and strict ideology that views all vampires with suspicion.

Sydney Sage, an eighteen-year-old Alchemist, is undercover as a student at Amberwood Preparatory School in Palm Springs, California, protecting Jill Mastrano Dragomir, a Moroi princess whose survival is politically necessary to keep the Moroi queen, Vasilisa Dragomir, on the throne. Sydney's team includes Eddie Castile, a dhampir guardian posing as her twin brother; Angeline Dawes, a dhampir from the Keepers, an isolated community of humans, dhampirs, and Moroi; and Adrian Ivashkov, a Moroi who wields spirit, a rare and volatile form of Moroi magic.

The novel opens with Sydney traveling to a Virginia bunker to testify about Keith Darnell, a corrupt former Alchemist she helped arrest for trafficking vampire blood. Keith desperately begs not to be returned to Re-education, an Alchemist facility for those deemed too sympathetic to vampires. Sydney testifies that Keith is selfish but not dangerously attached to vampires; the Alchemists send him back anyway. A guard shows Sydney a cell containing Liam, a human who served Strigoi and yearns to become one, reinforcing Alchemist warnings about vampire influence. The visit leaves Sydney conflicted: She is praised as a model Alchemist, yet she privately acknowledges she has grown to care about the vampires and dhampirs she works with.

Back in Palm Springs, Sydney manages the group's cover, handles Angeline's disciplinary problems, and supports research led by Sonya Karp, a Moroi restored from being a Strigoi through spirit magic, and Dimitri Belikov, a dhampir who underwent the same restoration. They are trying to discover why restored Strigoi appear immune to being turned again.

Sydney's classmate Trey Juarez introduces her to Brayden Cartwright, a brainy public school student who works as a barista at Spencer's, a local coffee shop. They bond over intellectual interests and begin dating, but Sydney feels no romantic spark. Meanwhile, her teacher Ms. Terwilliger, secretly a witch, pressures Sydney to develop her aptitude for human magic. Sydney resists, believing magic is wrong for humans.

A man named Jeff Eubanks, later revealed to be one of the vampire hunters, approaches Sonya, claiming to know her from Kentucky, where she had been a Strigoi. Shortly after, Sonya and Sydney are ambushed in an alley by four men. One holds a sword engraved with archaic alchemical symbols to Sonya's throat and tells her to "go back to Hell." Dimitri fights them off. The sword's symbols connect to a medieval faction that once shared origins with the Alchemists. Feeling helpless, Sydney enrolls in a self-defense class with Adrian, taught by Malachi Wolfe, an eccentric former survivalist whose philosophy of awareness and avoidance empowers her.

At Clarence Donahue's house, where the elderly Moroi hosts blood feedings, Sonya asks for a sample of Sydney's blood for research, noting that Strigoi were unable to drink it during a previous attack on Sydney. Sydney refuses, panicking at the idea of vampires having her blood. Adrian fiercely defends her, and Sydney realizes he understands her deep-seated fears better than anyone.

Sydney drives Adrian to San Diego to visit his father, Nathan Ivashkov. Nathan dismisses Adrian's efforts, blames him for his mother's imprisonment, and tears down his character. Sydney defends Adrian, but Nathan is unmoved. On the drive home, she stops for cherry slushes, secretly drinking one so Adrian will not interpret the gesture as pity. She later arranges for Queen Vasilisa to commend Adrian to his father, a kindness he eventually discovers and is deeply moved by.

Two men confront Sydney at a café, identifying themselves as the Warriors of Light, an organized group of vampire hunters with ancient ties to the Alchemists. They demand Sonya's location, believing she is still a Strigoi. Sydney refuses. At Clarence's house, the elderly Moroi confirms the hunters' existence and mentions a human named Marcus Finch who once saved him from rogue Warriors.

Sydney reluctantly gives Sonya a blood sample. Results reveal traces of elemental magic, which Sydney attributes to her tattoo. When Sonya pushes for more samples and permission for a Moroi to taste the blood, Adrian angrily intervenes.

At the Halloween dance, Adrian arrives needing a ride home. In his candlelit apartment during a power outage, he confesses his fear of spirit-induced insanity and his gratitude for Sydney's behind-the-scenes help with his father. Sydney gives him her gold cross necklace as a grounding object against spirit's destabilizing effects. Adrian nearly kisses her but is interrupted when the power returns. He tells her Brayden should have called her "the most beautiful creature I have ever seen walking this earth."

When Sonya disappears, Sydney suspects the Warriors. Overcoming her resistance to magic, she performs a scrying spell, a form of magical divination, that reveals a compound bearing a sun symbol identical to a tattoo on Trey's back. Trey admits the Warriors have Sonya and are holding combat trials to determine who will execute her. Sydney negotiates entry to the compound to plead Sonya's case, while Dimitri secretly coordinates a guardian rescue and Eddie plants a tracking device in Sydney's purse.

At the compound, Sydney watches Trey deliberately lose his fight, unwilling to win the right to kill. Sonya is dragged to a decapitation block, and Sydney argues before the council that Sonya was restored through spirit and is now an innocent Moroi. The council dismisses her plea. Before the sword falls, guardians storm in with tranquilizer guns. Dimitri, Eddie, and Angeline free Sonya while Sydney uses defensive spells she secretly prepared to escape. Adrian, who followed the rescue team against orders, finds her on the compound's perimeter using his ability to see her aura, or energy field, in the dark. They flee in his Mustang.

In the aftermath, Trey and his father are exiled from the Warriors for enabling the rescue. Donna Stanton, Sydney's Alchemist superior, confirms the Alchemists knew about the Warriors and praises Sydney's actions. Brayden breaks up with Sydney, which amuses rather than upsets her. Clarence gives Sydney a photograph of Marcus Finch, revealing Marcus has a modified Alchemist tattoo, though Stanton claims no record of him exists. Sydney resolves to find Marcus and learn the truth.

Sydney visits Adrian to confront him about faking his inability to drive. He admits he created excuses to be near her, then pulls her close and kisses her. Unlike her passionless kisses with Brayden, this one overwhelms her entirely. Adrian confesses he has fallen in love with her, calling her "light made into flesh" and saying she inspires him to be better. Sydney rejects him, citing the prohibition against human-vampire relationships and the golden lily tattoo as a vow she cannot break. She walks away without looking back, her heart breaking even as the lily on her cheek reminds her of who she is supposed to be.
